On the test 3 and test 4 , we added 5 drops of isopropyl alcohol into both 3 small test tubes and to investigate the differences of enzyme affecting the visibility of DNA. As you can see from the observation above, 3 drops of isopropyl alcohol did not show much of a difference into extracting a large scale of DNA compared to dropping of 5 drops of isopropyl alcohol. In our group we did 2 test on how enzyme affects the DNA.
The figure 1. 3 and figure 1. 4 shows how clearly meat tenderizer with isopropyl alcohol performs a better enzyme compared to pineapple juice. It breaks down the nucleus to release DNA better than pineapple juice. On the first test we actually did 3 drops of isopropyl alcohol, we then see the result it was difficult to distinguish between the egg yolk and the DNA. On third test, we added 5 drops of isopropyl alcohol and the results came in few minutes, we are then able to see the DNA clearly as white stringy substance appears. On the second test we tried a different type of enzyme, pineapple juice. We started off with 3 drops of pineapple juice, few minutes later there is no difference to the egg yolk, then on test 2 we did on the 5 drops of pineapple juice and there are DNA but it is very hard to notice the white stringy substance of DNA.
Discussion : Source of Enzyme Meat Tenderizer Results Meat tenderizer acts as a catalyst to break down, it breaks the open the cell nucleus to release the DNA. The juice of the pineapple contains Bromelain , a substance which able to break down protein into amino acid. Which DNA extraction the best?
DNA extractions can come in many different ways and forms by using different materials, enzyme, alcohol, food, you will get different results, all of them are important to play a big role of getting white springy substance out of the food. For instance our group had two different enzyme, meat tenderizer and alcohol, we did both of them in a fixed experiment 3 drops of isopropyl on meat tenderizer then 3 drops of isopropyl in pineapple juice, we got different result but concentration is fixed. We compared our results with group B and we got different results as well.
As group B used different enzyme and different concentration but as we can see from group B’s result, it tells us that the group B did use the isopropyl enzyme and by using that they get the most visible DNA out of it. Therefore this can also relate as, concentration makes the results different, as we used high amount of concentration we get high amount of DNA visibility because all of these white participate are separated by alcohol. But this does not mean only concentration play a role but as well as enzyme from our results.
Again I can conclude that our group had a better DNA extraction over other group because we fixed alcohol as our variable and manipulated enzyme as another factor, this can only relate to the other group result by concentration. 2. How are you going to compare the amount of DNA you extract? The amount of DNA extracted is seen by the thickness of the white springy substance the more of it means the more DNA has been extracted out because enzyme breaks nucleus and gets DNA out of it and alcohol precipitates DNA and separation takes place. Therefore if you can see white springy stuff it actually means many DNA has been tangled up. . Does the source of tissue (Eg fresh,frozen, cooked) make a difference? The source of tissue as well does affect the amount of enzyme extracted. The table shows the results of Group C explained about source of tissue make a difference. The tissue shows in frozen food can be seen much than others because when the temperature is low it does not allow DNA to have much activity so when it is cold the DNA works at a slow speed.
So its easier for enzyme to break nucleus and extract the DNA out. Tan 1 4. Do different method makes a difference? As we compare results with group B, methods does make a difference, the enzyme and concentration of alcohol can get you different results of DNA extracted. Our group did isopropyl alcohol and we compared with Group B’s one result using isopropyl we did get the almost the same amount of visibility. But as seen from other results like acetone as alcohol shows a different result as visibility is not that visible.
This again proves that using different method makes a difference for example concentration 5. Which approach gave the best result? The one with higher concentration and the enzyme will give you better results over the one which has low concentration of alcohol and weaker enzyme working to break the nucleus. For high concentration alcohol would be isopropyl alcohol and good enzyme that breaks the nucleus for our group would be the meat tenderizer. 6. How does your methods compare with methods currently used in research.
Our method we did in science laboratory is just the surface of genetic engineering. The point of doing the experiment was to extract out DNA for our education purpose and to compare with other groups factors causing DNA extraction differences. Generally, DNA extraction is a process for researchers/scientist to get DNA from nucleus for the work purpose, to diagnosed on humans health field, to study the gene, to copy or clone them, these can help save lives of other people. DNA is also for research on plants and animal to see how their body works to see how they actually grow by looking at the gene.
